Hello!

I'm mostly use my Blackstar FLY 3 with batteries but sometimes I use it with power supply. So I wonder: do I need to remove batteries every time when using power supply?

No, but it's good practice. I wouldn't leave batteries in for an extended period of time. Possible acid leak and/or just running them down.
